Deoxyribosyl Transfer. I. Thymidine Phosphorylase and Nucleoside Deoxyribosyltransferase in Normal and Malignant Tissues.

چکیده
Nucleoside : purine (pyrimidine) deoxyribosyltransferase activity (EC 2.4.2.6.) has been demonstrated in bacteria by a number of investigators (l-5). Similar activity in mammalian tissues was reported by deVerdier and Potter (6). With the mammalian enzyme, however, the substrate specificity of the transfer reaction appeared to be restricted to pyrimidine bases and their deoxyribonucleosides. Transferase activity was demonstrated in normal rat liver but not in the Novikoff hepatoma or a number of other rat tissues. Since the Novikoff hepatoma has thymidine phosphorylase activity (EC 2.4.2.4. ) which catalyzes Reaction 1, the absence of Reaction 2, catalyzed by the transferase, was not expected since reversal of phosphorolysis should theoretically result in equilibration between the labeled pyrimidine base and the pyrimidine of the nucleoside.

متن کاملRibosyl and deoxyribosyl transfer by bacterial enzyme systems.
The enzymatic transfer of ribose and deoxyribose residues in pyrimidine nucleosides to purines was catalyzed by cell-free extracts of various bacteria. Almost all the strains belonging to Enterobacteriaceae were capable of catalyzing the transfer reactions. The transfer activities were also detected among some bacterial strains of other families: Pseudomonadaceae, Corynebacteriaceae, Micrococca...

Several types of deoxyribosyl compounds have been found in the acidsoluble fraction of normal and malignant tissues (l-8). In 1955 Potter and Schlesinger (1) reported the isolation of deoxynucleoside mono-, di-, and triphosphates from extracts of calf thymus. More recently LePage (7) reported the isolation of deoxyadenosine triphosphate from a tumor. Schneider (2), using a microbiological assay...
متن کاملAcquisition of human concentrative nucleoside transporter 2 (hcnt2) activity by gene transfer confers sensitivity to fluoropyrimidine nucleosides in drug-resistant leukemia cells.
CEM-ARAC leukemia cells with resistance to cytarabine were shown to lack equilibrative transporter (hENT1) expression and activity. Stable transfer of hCNT2 cDNA into CEM-ARAC enabled Na(+)-dependent transport of purine and pyrimidine nucleoside analogs and provided a unique in vitro model for studying hCNT2. Analysis of [(3)H]uridine inhibitory activity by test substances in hCNT2 transfectant...
متن کاملThe activity of thymidine phosphorylase correlates with tumor size and lymph nodes status in breast carcinoma.
The platelet-derived endothelial cell growth factor (PD-ECGF) is one of the potent angiogenic factors. Recently, its homology with thymidine phosphorylase (dThdPase), an enzyme involved in pyrimidine nucleoside metabolism, has been shown. In the present study, dThdPase activity was evaluated spectrophotometrically in 43 breast carcinomas and in 19 cases of non-neoplastic breast tissues. The mea...
